Inhibition by Nucleosides

چکیده
FUdR is readily phosphorolyzed by thymidine phosphorylase (EC 2.4.2.4) purified from Escherichia coli (22), and it also can be cleaved by uridine phosphorylase (EC 2.4.2.3) purified from Ehrlich ascite tumor (21). It has been suggested that this breakdown may be a reason for the limited therapeutic effect of FUdR, and various attempts to find conditions that prevent phosphorolysis of FUdR have failed (2, 8, 12, 13). It has been known for a while that in bacteria various nucleosides can prevent the phosphorolysis of thymidine in vivo and thus promote its incorporation into cellular deoxyribonucleic acid (3, 4, 6, 16, 18, 25). In the present report we have investigated whether in E. coli the cleavage of FUdR can be prevented by nucleosides and whether uridine phosphorylase of E. coli can catalyze this phosphorolytic reaction as is the case in mammalian cells.

Stannous Chloride: A Reagent for Removal of Dimethoxytrityl Group from 5'-Dimethoxytrityl Nucleosides
Detritylation of 5'-dimethoxytrityl nucleosides have been quantitatively achieved in minutes at room temperature under aprotic neutral conditions by using stannous chloride. Of additional practical consideration and in contrast to protic acids, no depurination was observed with this reagent.

The glycosylation of 1,3,4-oxadiazole-2-thiones has been performed with peracetylated β-pyranosyl bromide in the presence of potassium carbonate. Deprotection of acetylated thioglycosides was necessary for increasing their antibacterial effects. The structure of nucleosides were confirmed by 1H NMR, 13C NMR and HRMS. The anomeric protons of nucleosides c1–4 were assigned to the doublet, confirm...

The interaction of nucleosides with the glucose carrier of human erythrocytes was examined by studying the effect of nucleosides on reversible cytochalasin B-binding activity and glucose transport. Adenosine, inosine and thymidine were more potent inhibitors of cytochalasin B binding to human erythrocyte membranes than was D-glucose [IC50 (concentration causing 50% inhibition) values of 10, 24,...
